Article The Effects of Ovarian Vitrification on Mitochondrial Fusion (mfn-1, Mfn 2 and Opa-1), Fission (dnm-1), Mitophagy (parkin, Pink-1) Andtransport (miro-1, Milton) Proteins(2020) Vatansever, H. Seda; Özbilgin, Kemal; Öztatlıcı, Mustafa; Temel, Merve; İnan, SevinçObjective: Ovarian cryopreservation is a useful alternative for fertility preservation in assisted reproductivetechnologies. In spite of many advances in the vitrification procedure, this technique is still considered experimental. Therefore in this study, we aimed to investigate the expressions of mitochondrial fusion (MFN1, MFN2 and OPA1), fission (DRP1), mitophagy (PARKIN, PINK1) and transport (MIRO-1, MILTON) proteins in ovarian tissues by qPCR technique after vitrification.Materials and Methods: To investigate the mitochondrial dynamics after vitrification, the ovaries were recoveredfrom 6-8 week old healthy female mice (No: 12) and were divided into vitrification and control groups. Vitrification carried out using ethylene glycol, dimethylsulfoxide and sucrose. After total RNA isolation from ovaries in control and vitrification groups, qPCR technique was performed to determine the expression rate of target genes. The relative gene expressions of the target genes were evaluated according to 2???Ct method.Results: Histological evaluation revealed that ovaries in the control group were shown normal morphology while the tissue integrity of the ovaries in the vitrification group is disrupted, some follicles are degenerated and granulosa cells were shed into antrum. According to our qPCR results, outer membran fusion proteins MFN1 gene expression decreased 1.12 fold and inner membran protein OPA-1 increased 1,36 fold in the vitrification group compared the control group. The mitochondrial fission protein DRP-1 gene expression increased 1.20 fold in the vitrification group. The mitophagy proteins PINK-1 and PARKIN genes expressions decreased 1.34 and 3.75 fold respectively in the vitrification group. The transport proteins; MIRO-1 gene expression decreased 1.16 fold but MILTON (TRAK-1) gene expression sharply increased 2,28 fold compared the control group.Conclusion: The alternation of the mitochondrial dynamics related gene expressions may lead a decrease in themitochondrial function during the ovarian vitrification and may reduce the potential of oocyte maturation and embryo development.Other Evaluation of Jak/Stat Signaling Pathway-Associated Protein Expression at Implantation Period: an Immunohistochemical Study in Rats(2022) Gürel, Çevik; Kuşçu, Gökçe Ceren; Vatansever, H. Seda; Köse, Can; İnan, Sevinç; Özbilgin, Kemal; Kırmaz, CengizObjective: The implantation period of gestation is a complex process in which numerous molecular pathways play a role. The Janus kinase/signal transducers and activators of transcription (JAK/STAT) pathway is one of the evolutionarily conserved cascades used to transduce a multitude signals for several biological events such as implantation and uterine receptivity. Previous studies have indicated that the implantation process is disrupted by the lack of proteins involved in this pathway. However, our literature knowledge showed that there is no study evaluated the expression of JAK/STAT signaling pathwayassociated proteins during the implantation period. This study investigated the expression patterns of JAK/STAT signaling pathway-associated proteins in rats by immunohistochemical (IHC) staining according to gestational days. Methods: In this study, thirty Wistar Albino rats weighing 250-300 g, without any problems in their menstrual cycles, were used. The pregnant animals were sacrificed on the 4th, 5th, and 6th days and histochemical and IHC analysis were performed on the uterine tissues taken from these animals. Results: In this study, protein expression of JAK1, JAK2, JAK3, STAT2, STAT3, STAT4, STAT5, and STAT6 belonging to the JAK/STAT pathway was evaluated in the uterine surface epithelium, gland structures, antimesometrial region, cells of the immune system, myometrium, mesometrial region and decidual cells, which are associated with the implantation process. The result of this evaluation reveals that the expression levels of these proteins in the JAK/STAT pathway vary in different days of the implantation period in implantation-related structures. Conclusion: This study indicates that JAK/STAT signaling pathway-associated proteins can function actively in the regulation of the immunological response of the uterus and embryo-uterus interaction during the implantation period in rats. Seda; Onal, Tuna; Kutlu, Necip; Ozel, Fehmi; Gunlusoy, Bulent; Gumus, Bilal H.Aim The dorsal nerve of the penis (DNP) is the terminal branch of the pudendal nerve which is responsible for the somatic innervation of the penis. This study aims to outline any direct role of the DNP in the hemodynamics of erection histologically and physiologically. Materials and Methods Fifteen Wistar albino rats were sorted into the electrical activity (n = 6), intracavernous pressure (n = 4), and control (n = 5) groups. The dorsal nerve was electrostimulated and the simultaneous changes in intracavernous pressure and smooth muscle activity were recorded. Penile tissues were collected, fixed, and sectioned, the slides were stained with either hematoxylin-eosin for morphological evaluation or using the indirect immunoperoxidase technique to analyze the distributions of eNOS, iNOS, and nNOS. Results During electrostimulation, there was a simultaneous statistically significant decrease in the electrical activity inside the corpora in electromyography and an increase in intracavernous pressure. eNOS and iNOS immunoreactivities were higher in the study group than in the control group. nNOS immunoreactivity was moderate in both study and control groups. Conclusion Some fibers in the dorsal nerve of penis continue into the corpora cavernosa through the tunica albuginea and have an active, direct role in the hemodynamic process of erection, which may be complementary to the main route of innervation.Article Meme Kanseri Hücre Hatlarında Propranolol ve Paklitakselin Anjiyogenez Üzerine Etkisi(2019) Vatansever, H. Seda; Tuğlu, İbrahim; Özbilgin, Kemal; İnan, Sevinç; Şimşek, Fatma; Müftüoğlu, SevdaAmaç: Bu çalışmanın amacı infantil hemanjiyomvakalarında kullanılan propranolol (PR) ile kemoterapötikbir ajan olarak yaygın kullanılan Paklitakselin (PX) kanserhücreleri üzerine etkisini incelenmesidir.Gereç ve Yöntem: Tripan mavisi ile hücre sayımıyapılarak hücrelerin ikilenme zamanları belirlendi. MTTtesti ile de ilaçların sitotoksik etkisi ve IC50 değerleri analizedildi. İnvazyon yönünden farklı iki meme kanseri hücrehattında (MDA-MB-231 ve MCF-7) anti-VEGF, antieNOS, anti-iNOS ve anti-ERK1/2 primer antikorlarıindirek immunohistokimyasal yöntemle incelendi.İmmunoreaktivitenin değerlendirilmesi için H skorlamasistemi kullanıldı.Bulgular: MTT testi ile hücrelere uygulanacak ilaçdozlarının IC50 değerleri MDA için; PX: 5 nmol, PR: 50µm ve MCF- 7 için; PX: 3,7 nmol, PR: 50 µm olarakbulundu. İmmunohistokimyasal uygulamada kanserhücrelerinde kontrol gruplarının immunoreaktivitesişiddetli ve/veya çok şiddetli artmış iken PX, PR vekombine uygulanan ilaç gruplarında boyanma şiddetianlamlı veya çok anlamlı olarak azaldı.Sonuç: Bu çalışma ile kemoterapötik olarak uygulananpaklitaksele ek olarak anti anjiyogenik ilaç uygulamalarınındamarlarda vazodilatasyon, hücre çoğalması, göçü veyaşam süresini etkilemesi sonucunda anjiyogeneziazaltması veya önlemesi açısından meme kanserinintedavisinde önemli olduğu düşünülmüştür.
