    Protective Effects of Taurine Against Renal Ischemia/Reperfusion Injury in Rats by Inhibition of Gelatinases, Mmp-2 and Mmp-9, and P38 Mitogen-Activated Protein Kinase Signaling

    Dysregulated expression of matrix metalloproteinases (MMPs) is closely associated with the pathogenesis of renal ischemia/reperfusion injury (I/R). The production of excessive reactive oxygen species (ROS) causes tissue damage. Increased ROS production causes activation of p38 mitogen-activated protein kinase (MAPK) signaling, which participates in gene regulation of MMPs, especially MMP-2 and MMP-9 (gelatinases). Taurine (2-aminoethanesulfonic acid) in mammalian cells functions in bile acid conjugation, maintenance of calcium homeostasis, osmoregulation, membrane stabilization, and antioxidation, antiinflammatory, and antiapoptotic action. We investigated the effects of taurine and the possible role of p38 MAPK signaling on regulation of MMP-2 and MMP-9 in a renal I/R injury model in rats. Rats were divided into three groups: sham, I/R, and I/R + taurine treated. After a right nephrectomy, I/R was induced by clamping the left renal pedicle for 1 h followed by 6 h reperfusion. Taurine was administered 45 min prior to induction of ischemia. Renal function was assessed by serum creatinine and blood urea nitrogen (BUN) levels. Tubule injury and structural changes were evaluated by light microscopy. Malondialdehyde (MDA) levels were analyzed by high performance liquid chromatography (HPLC). Superoxide dismutase (SOD) activity levels were measured using a colorimetric kit. mRNA expression of MMP-2 and MMP-9 was determined by real-time polymerase chain reaction. MMP-2 and MMP-9 activities were measured using a fluorimetric kit. Phosphorylated p38 (p-p38) and total p38 MAPK protein expressions were evaluated by western blot. Taurine pretreatment significantly attenuated renal dysfunction and histologic damage, such as renal tubule dilation and loss of brush borders. The pretreatment also decreased the MDA level and attenuated the reduction of SOD activity in the kidney during I/R. Taurine pretreatment also decreased significantly both MMP-2 and MMP-9 mRNA expression and MMP-9 activity induced by I/R. In addition, the activity of p38 MAPK signaling was down-regulated significantly by taurine administration. Inhibition of MMP-2 and MMP-9 expression and MMP-9 activity caused by taurine may be associated with suppression of p38 MAPK activation during I/R induced renal injury in rats. Therefore, taurine administration may prove to be a strategy for attenuating renal I/R injury.

    İnsan Primer Fibroblast Hücre Kültür Karakterizasyonu İçin Biyosensör Geliştirilmesi

    Hücre Kültürü, deneysel çalışmaların vazgeçilmez araçlarından biridir. Üç boyutlu organizmanın doğrudan kullanılamadığı pek çok deneysel çalışmanın uygulanabilir modeli olarak tercih edilmektedir. Herhangi bir doku parçası mekanik ya da enzimatik yollarla belirli hücreler elde etmek için kültüre edilebilir. Bu kültür işlerinin inceliklerinin yanında, primer kültürlerde ayrıca dikkat edilmesi gereken en önemli nokta, heterojen bir hücre topluluğunun içinden kendi ilgilendiğimiz hücre topluluğunun karakterizasyonunu yaparak deneylere bu hücreler üzerinden devam edebilmektir. Bu projede, ticari olarak elde edilebilen hücre hattının, ya da stabil ve kanıtlanmış bir deney hayvanı modelinin bulunmadığı durumlarda sıklıkla tercih edilen ve insandan doğrudan elde edilen primer kültürler üzerinde durulmuştur. Bu projenin amacı; çeşitli dokulardan elde edilen primer fibroblast hücre kültürlerinin karakterizasyonu için fibroblast biyobelirteci olarak bilinen ısı şok proteini 47 (HSP47) proteinini tanıyan bir biyosensör geliştirilmesinin sağlanmasıdır. Biyosensörler, analit ve biyoaktif molekül ilişkisine dayalı olarak geliştirilen ucuz, güvenilir ve hızlı ölçüm sistemleridir. HSP47 biyosensörünün geliştirilmesi için, spesifik moleküller olan antikorların, dönüştürücü yüzeyinin modifikasyonundan sonra yüzeye immobilize edilmesi ve bundan sonra da HSP-47 molekülü ile antikor arasındaki afiniteye bağlı olarak yüzeyin elektrokimyasal olarak incelenmesi gerçekleştirilmiştir. Bunun için altın nanopartikül elektrotlar kullanılmış ve grafen oksit ile modifiye edilmişlerdir. Biyosensörün performansı hem standartlarda hem de hücrelerde test edilmiştir. Biyosensörün düşük tespit limiti (LOD) ve lineer tespit limiti (LOQ) sırasıyla 0,06 ng/mL ve 0,183 ng/mL olarak hesaplandı. Doğrusal ölçüm aralığı 10-160 pg/mL arasında elde edildi. Böylece literatürde ilk kez HSP47?nin primer fibroblast kültürlerinde belirlenmesi için impedimetrik bir biyosensör geliştirilmiş oldu.

    İşletmeler dinamik yapıdaki kurum ve kuruluşlar oldukları için her geçen gün değişmekte ve bu değişime ayak uydurarak ayakta kalabilmektedirler. Bu değişim sürecini etkin şekilde kontrol eden mekanizma kalite standartlarıdır. Kalite standartları, işletmelerin rekabet ortamındaki konumlarını ele alırken aynı zamanda toplumdaki varlıklarını geliştirmek ve iyi bir hizmet sunmak adına da işletmeleri sosyal, toplumsal, üretim biçimi vb. ölçütlerde düzenleme gayesi güderler. Bu gaye doğrultusunda oluşturulan standartlardan biri de SA 8000 standardıdır. CEPAA tarafından 1997 senesinde oluşturulan bu standart, işletmelere ilişkin etik değerlendirmeler getirmesi yönüyle diğer standartlardan ayrılmaktadır. Bu çalışmada SA 8000 standardı, sağlık kuruluşlarındaki rolüyle ele alınmış ve hasta güvenliğiyle ilişkisi sorgulanmıştır. Bu kapsamda çalışmada Türkiye'de 2020 senesine değin hazırlanan Yüksek Lisans çalışmalar değerlendirilmiş; sağlıkta kalite, hasta güvenliği, sosyal sorumluluk ve SA 8000 anahtar kelimelerinden yola çıkarak yapılan bibliyometrik analiz ile birlikte SA 8000'in hasta güvenliğiyle ilişkisi değerlendirilmiştir.

    In Vitro Effects of Selenium on Human Glioblastoma Multiforme Cell Lines: a Preliminary Study

    Glioblastoma multiforme (GBM) is caused by the central nervous system-derived glial cells, and represents the most common (50%-60%) form of primary brain tumors. The aim of this study was to investigate the in vitro effects of selenium on human GBM cells. In the present study, GMS-10 and DBTRG-05MG human GBM cell lines were used as a model to examine selenium entering the cell, cell proliferation, cytotoxicity, DNA fragmentation and Ki-67 protein expression in selenomethionine treated and non-treated groups. Seleno-L-methionine (SeMet) as the organic source of selenium exerted effects on cell proliferation and cytotoxicity, as assessed with WST-1 and lactate dehydrogenase (LDH) tests, respectively. Apoptosis was assessed by DNA fragmentation with an enzyme-linked immunosorbent assay. Ki-67 protein expression was determined by Western blotting, while selenium measurements were performed in the supernatants and lysates by using Graphite Furnace Atomic Absorption Spectrometry. This is the first study to examine the effects of SeMet on cell proliferation and death in GMS-10 and DBTRG-05MG cells. Both GBM cell lines responded to SeMet in a dose- and time-dependent manner. WST-1 test showed that low-dose SeMet treatment (50 and 100 mu M) increased cell proliferation. Analysis of intracellular SeMet levels by using AAS showed results consistent with viability and cytotoxicity tests. SeMet treatment for 72 h caused increased DNA fragmentation in both cell lines. In conclusion, our results suggest that SeMet induces cell death at high doses, while increasing cell proliferation at low doses. In the view of the data obtained in this investigation, further studies focusing on the possibility of using SeMet against different types of GBM and in combination with prospect synergic compounds are considered to be worthwhile.

    The New Horizons in Biochemistry and Molecular Biology Education Conference was organized by the International Union of Biochemistry and Molecular Biology (IUBMB) in collaboration with the Federation of European Biochemical Societies (FEBS), and the Weizmann Institute of Science (Israel) and held in Rehovot, Israel, on September 6-8, 2017. The program covered the entire lifespan of students/scientists from the school level to undergraduate, graduate, and post-doctoral levels and brought together 130 international participants. This article provides an overview of the major issues and topics discussed at the conference and suggestions for the way forward.

    The Role of Resveratrol on Full - Thickness Uterine Wound Healing in Rats

    Objective: Healing of the uterus after cesarean section and myomectomy operation is clinically important. In this study, we aimed to investigate the effects of resveratrol (3,5,4'-o-trihydroxystilbene) on the wound healing process of the uterus in rats treated with resveratrol following full thickness injury of the uterus. Materials and methods: Twenty-one female wistar albino rats were divided randomly into three groups (1) control group with no intervention (2) injury group with uterine full thickness injury (3) resveratrol group with uterine full thickness injury and treated with resveratrol. Resveratrol was injected by oral gavage at the doses of 0.5 mg/kg/day for 30 days following uterine full thickness injury. Vascular endothelial growth factor (VEGF) and platelet-derived growth factor (PDGF) distributions were assessed using the immunohistochemical methods in tissue and ELISA methods in the tissue homogenate. Glutathione peroxidase (GPx) and superoxide dismutase (SOD) activities were evaluated with colorimetric method and malondialdehyde (MDA) levels also were measured using high performance liquid chromatography in the tissue homogenate. The effects of resveratrol on the uterine histology also were evaluated histologically with the light microscopy. Results: Histological evaluation and immunohistochemical evaluations showed that treatment with a resveratrol significantly increased the thickness of the uterine wall and VEGF expression and decreased expression PDGF during wound healing. Biochemically, GPx and SOD activities were increased significantly after treatment with resveratrol. Additionally, resveratrol administration decreased MDA levels. Conclusion: These results showed that the antioxidant effects of resveratrol has been shown to have a positive influence on wound healing of the uterus.
