Article 1,25-Dihydroxyvitamin D3 Induces N-Myc Downstream Regulated Gene-2 Expression in Papillary Thyroid Carcinoma Cells(Dokuz Eylul Univ Inst Health Sciences, 2020) Sipahi, Murat; Bartik, Didem Keles; Doruk, Mehmet; Bayraktar, Firat; Oktay, GulgunPurpose: In addition to its role in serum calcium homeostasis, the anti-tumor function of 1,25-dihydroxyvitamin D-3 (calcitriol) in cancer development is well established. N-myc Downstream Regulated Gene 2 which functions as a tumor suppressor gene has recently been shown to be downregulated in various cancer leading to increased tumor incidence, progression and metastasis. The goal of this study was to investigate the possible effects of calcitriol treatment on NDRG2 expression in BCPAP papillary thyroid carcinoma cells. Methods: The experiments were carried on human primary thyroid follicular epithelial cells (Nthy-ori-3-1), and human papillary thyroid carcinoma cells (BCPAP). The half maximal inhibitory concentration (IC 50) of calcitriol on BCPAP cells was determined by WST-1 assay. BCPAP cells were treated with 15 and 30 mu M calcitriol for 24, 48, and 72 hours, respectively. Basal NDGR2 expression in Nthy-ori-3-1 and BCPAP cells as well as the alterations on NDRG2 expression in calcitriol treated BCPAP cells were evaluated with western blot. Results: A significant downregulation of NDRG2 was observed in BCPAP cells when compared to Nthy-ori-3-1 cells (p<0.01). IC50 dose of calcitriol was found to be 64, 54 and 43 mu M for 24, 48 and 72 hours, respectively. NDRG2 protein expression levels were significantly increased in 30 mu M calcitriol treated BCPAP cells after 48 hours (p<0.05). Conclusions: Calcitriol induced NDRG2 protein expression in BCPAP cells. We predict that calcitriol increased NDRG2 protein levels in BCPAP cells via c-Myc repression, which is upregulated by aberrant Wnt/beta-catenin signaling. Methods: Eighty-eight female patients with SjS and 3,338 age-matched healthy female controls were included in this study. 25 OH-D3 levels were compared with healthy controls. Then the patients were stratified according to their 25 OH-D3 levels, either insufficient/deficient or normal (<50 nmol/L or ≥50 nmol/L). The disease activity was evaluated using The EULAR SjS disease activity index (ESSDAI) and its components. Correlation analyses were also performed for a possible correlation with disease characteristics and markers of activity. Results: No differences in 25 OH-D3 levels were found between SjS and healthy populations (p>0.05). No correla- tions were found between patient characteristics or labo- ratory values (p>0.05). Conclusions: This study did not find a link between disease characteristics and disease activity and 25 OH-D3 levels. The aim of the study is to investigate the effects of aconitine on lateral migration, and matrix metalloproteinase (MMP) activity in MDA-MB-231 triple-negative breast cancer cells. Material and Methods: A WST-1 viability assay was conducted to determine the effect of aconitine on the viability of MDA-MB-231 cells. Following treatment with non-cytotoxic doses of aconitine, lateral migration was evaluated through wound healing assays. Additionally, gelatin zymography was conducted to analyze MMP-2 and MMP-9 activity and secretion levels. Results: Aconitine concentrations up to 200 mu M did not significantly affect cell viability for up to 72 hours, whereas higher doses (400-600 mu M) reduced viability in a time-dependent manner. Aconitine at 200 mu M showed a trend towards decreased lateral motility, with a significant reduction at 9 hours post-treatment. Gelatin zymography revealed no alterations in MMP-2 and MMP-9 activity or secretion levels following aconitine treatment. Conclusion: Aconitine demonstrates limited efficacy in modulating the migratory capacity of MDA-MB231 cells and does not affect gelatinase activity. Although these vulnerabilities are often targeted individually, targeting all with an enzyme may prove a novel approach. However, therapeutic enzymes are prone to proteolytic degradation and neutralizing antibodies leading to a reduced half-life and effectiveness. We hypothesized that glucose oxidase (GOX) enzyme that catalyzes oxidation of glucose and production of hydrogen peroxide, may hit all these targets by depleting glucose; crippling anabolic pathways and producing reactive oxygen species (ROS); unbalancing redox homeostasis. Methods: We encapsulated GOX in an acrylamide layer and then performed activity assays in denaturizing settings to determine protection provided by encapsulation. Afterwards, we tested the effects of encapsulated (enGOX) and free (fGOX) enzyme on MCF-7 breast cancer cells. Results: GOX preserved 70% of its activity following encapsulation. When fGOX and enGOX treated with guanidinium chloride, fGOX lost approximately 72% of its activity, while enGOX only lost 30%. Both forms demonstrated remarkable resilience against degradation by proteinase K and inhibited viability of MCF-7 cells in an activity-dependent manner. Conclusions: Encapsulation provided protection to GOX against denaturation without reducing its activity, which would prolong half-life of the enzyme when administered intravenously.Article Citation - WoS: 2Citation - Scopus: 1Adjuvant Effects of Chemotherapeutics and Metformin on Mfe-319 Endometrial Carcinoma Cell Line(Editura Acad Romane, 2020) Aydemir, Isil; Uluer, Elgin Turkoz; Korkmaz, Oya; Tuglu, Mehmet Ibrahim; Inan, SevincWe aimed to investigate the cytotoxicity of Metformin, Cisplatin, and Paclitaxel on MFE-319 endometrial carcinoma cell line using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and immunocytochemistry assays. Half maximal inhibitory concentration (IC50) doses of three drugs alone and in the dual combinations were applied to the cells. Immunocytochemical method was performed for the cell survival and for phosphatidylinositol 3-kinase (PI3K), phosphorylated extracellular regulated kinases (pErk)-1/2, Akt-1, phosphorylated Akt (pAkt)-1/2/3 cell growth markers and angiogenic vascular endothelial growth factor (VEGF). Immunoreactivities were evaluated using H-score and analyzed using the one-way analysis of variance (ANOVA) test for statistics. It was found that these drugs caused a decrease in the immunoreactivities of these markers. Particularly, dual combination of Paclitaxel and Cisplatin decreased the immunoreactivities of PI3K, pErk-1/2, Akt-1, and pAkt-1/2/3. Cisplatin and Paclitaxel were more effective than Metformin; on the other hand, Metformin has been shown to enhance the efficacy of these two drugs. In vitro or in vivo further studies are needed to investigate the efficacy of these three drugs via PI3K/Akt signal pathway.Article Advertising Adaptations Between Globalization and Locality: Case of Snickers(Ankara Univ, Fac Communication, 2015) Kaptan, YesimBy comparing the American and the Turkish versions of television commercials arising from the Snickers' global advertising campaign You are not you when you are hungry, this article examines how commercials are adapted to appeal to a sense of locality through the strategies of locality. To explore how locality is represented in advertising, three localization strategies (national language, culture, humor) employed in advertising adaptation are analyzed. Using interviews with focus groups and an advertising practitioner, the author argues that advertising plays an important role for construction and reproduction of locality in everyday life. The author asks which values and codes are employed in advertising adaptations and how they are articulated to discourse of consumer culture in the media materials. The article also discusses the significance of advertising adaptations within the overlapping contexts of global formats, national media, and local identities.Article Aerospace 3d Textile Preform Analysis: Experimental and Mesomodeling(Polish Society of Composite Materials, 2023) Saboktakin, Abbasali; Safaei, H.For the purpose of predicting how textile preforms affect the quality of the composite material and its performance, the stitched textile preform must be characterized. Experimental compaction analysis and finite element analysis of textile preforms are the main subjects of this paper. The formability parameters of a preform can be changed by the stitching process, according to research on the mechanical properties of preforms conducted during compression testing. The load-deformation response, which is depicted in detail, had the greatest influence on preform deformation. Less fiber bundle undulation in the plane direction and more stitching thread undulation in the thickness direction were observed during compression of the stitched preform, whereas the stitching thread improved the resistance of the preform to compression loading. © 2023, Polish Society of Composite Materials. Methods: The study adopted a retrospective design. It was carried out on 208 kidney transplant recipients. The social cognitive theory was used for understanding and addressing the issues of nonadherence to immunosuppressive medication. Data were collected with a self-reported-Immunosuppressant Therapy Adherence Scale and biological assays. Descriptive analyses and logistic regression were used to analyze data. Results: According to the self-reported assessment, medication adherence of the patients receiving transplants from live donors was 0.503 times lower than that of the patients receiving transplants from deceased donors. The biological assays did not show any affecting factors in medication adherence. Conclusion: The results of the study showed the importance of combining methods to assess medication adherence. In the self-reported assessment, live donor transplantation was found to negatively affect medication adherence. Turkiye is the country with the highest number of kidney transplantations from live donors in the world. Simple recipes and procedures were determined for the adaptation and use by anyone who is interested to use sustainable, non-toxic, and even edible plastic-like materials without the need for a scientific background. The recipes and procedures were developed by controlled laboratory experiments. The main recipes contain different combinations of agar strips, water, glycerine, and vinegar. A variety of films with different sensorial properties (soft, hard, flexible, stiff clear, coloured, textured, with or without spicy odour, etc.) were prepared by heating the ingredients in a saucepan, mixing, pouring on a flat surface, and drying for a few days. These were evaluated by tinkering and user studies that involved students from design, food engineering, culinary arts, and other fields. As a proactive approach, lifelong learning presents an opportunity for the elderly to challenge stereotypes, remain engaged in society, and foster personal growth. This study highlights how training and educational programs can empower older adults, instilling a sense of purpose and accomplishment, leading to a more fulfilled and satisfying life in their later years. Bu amaçla, 37 maddelik bir bilgi ölçeği ve 23 maddelik birtutum ölçeği, İzmir’in farklı üniversitelerinde okuyan yaşları 17 ve üstü olan1025 öğrenciye kolay ulaşılabilir durum örneklemesi yöntemi kullanılarak testedilmiştir. Bilgi ölçeğinin madde ve faktör analizlerinden sonra ölçekten 16madde çıkarılmıştır. Oblik döndürme sonucu, varyansın %34’ünü açıklayanüç temel bileşen olduğunu göstermiştir. Bilgi ölçeğinin güvenirliğini sınamakiçin Kuder-Richardson-20 katsayısı hesaplanmış ve .76 olarak bulunmuştur.Tutum ölçeği için faktör analizi yapıldıktan sonra 6 madde çıkarılmıştır. Oblikdöndürme, varyansın %42.43’ünü açıklayan iki temel bileşeni ortaya çıkarmıştır.Ölçeğin Cronbach alfa değeri .90 olarak bulunmuştur. İki ölçek arasındaki ilişkiincelendiğinde ise, AIDS konusunda daha fazla bilgiye sahip kişilerin AIDS’ekarşı daha olumlu tutuma sahip olduğu gözlenmiştir. Yöntemler: Temmuz 2018 ile Ağustos 2019 tarihleri arasında aile hekimlerinden veri toplamak amacıyla çevrimiçi bir anket geliştirildi. Tanımlayıcı ve korelasyonel bir araştırma deseni kullanıldı. Çalışmada uygulanan anket, “katılımcıların kişisel bilgileri” ve “Sağlıklı Yaşam Biçimleri Davranış Ölçeği-II”i içermektedir. Kesitsel bir çalışma olarak tasarlanan bu araştırmada, örneklem kolayda örnekleme metodu ile sekiz ülkeden belirlenmiştir, ve anketler katılımcılara elektronik ortamda uygulanmıştır. Katılımcılara anketin süresi, veri saklama işlemleri, saklama süresi, araştırmacıların kimliği ve çalışmanın amacı hakkında bilgi verilmiştir. Bulgular: Çalışmaya 131 aile hekimi katılmış olup, erkek katılımcı oranı daha yüksek bulunmuştur. Sigara içme sıklığının Türk aile hekimleri arasında anlamlı derecede yüksek olduğu tespit edilmiştir (p=0,013). Ancak, ölçek toplam puanı veya altı boyutta alınan puanlar açısından aile hekimleri arasında istatistiksel olarak anlamlı bir fark bulunmamıştır. Sonuç: Çoğu aile hekimi, bu alanda daha fazla eğitim almak istediklerini belirtmiştir, bu da tıp fakültelerinin müfredatına eğitim müdahalelerin dahil edilmesi gerekliliğini göstermektedir. Ayrıca, iyi yapılandırılmış web tabanlı kitlesel çevrimiçi kurslar, aile hekimlerinin sağlıklı yaşam biçimi davranışları ve yaşam biçimi tıbbı uygulamalarına yönelik eğitim gereksinimlerini karşılamada etkili bir çözüm olabilir.Article Alternative E-Assessment in Language Classes: Recent Experiences and Future Directions(2024) Çağlayan, EsinThis study aimed to explore English as a Foreign Language (EFL) teachers' perceptions regarding alternative e-assessments in the Turkish tertiary level context. The study utilized semi-structured interviews with a sample of EFL instructors from 17 higher education institutions in Türkiye. The data were analyzed using thematic analysis to identify emerging themes. The results revealed varying perceptions about alternative e-assessments, with six primary themes emerging: impact on learning, workload, academic integrity, digital technology, grading and feedback, and assessment competence. Overall, this qualitative study provides valuable insights into the perceptions of EFL teachers regarding alternative e-assessments, highlighting key areas of concern. The study emphasizes the need to address these concerns in professional development programs that provide teachers with the necessary knowledge and skills to effectively implement alternative e-assessments. The results also have implications for the development of effective assessment policies and procedures, optimizing educational practices in the digital age.Article Alzheimer Hastaliği Klinik Olcutlerine Gore Demans ve Hafif Kognitif Bozukluk Ayriminda Sozel Akicilik Testlerinin Mini Mental Durum Testi ile Birlikte Kullanilmasi Yararli Olabilir(Cukurova Univ, Fac Medicine, 2022) Isbitiren, Yagmur Ozbek; Kiyi, Ilayda; Yener, GörsevPurpose: The aim of this study is to present the sensitivity, specificity and cut-off points of Mini Mental State Examination (MMSE), semantic and phonemic fluency tests determined by ROC curve analysis in differentiating between Alzheimer's dementia (AD) and mild cognitive impairment (MCI).Materials and Methods: Age-education-sex matched 72 healthy individuals, 72 MCI and 72 AD dementia individuals were included in the study. MMSE, semantic and phonemic fluency tests were applied to all participants. Sensitivity and specificity values and cut-off scores of MMSE, semantic and phonemic fluency tests in distinguishing healthy, MCI and ADD individuals were obtained by ROC curve analysis.Results: ROC analyses show that all three tests can significantly discriminate between healthy, ADD and MCI individuals. It was seen that the discrimination power of the MMSE was higher than the verbal fluency tests when the areas under the ROC curve were compared. The specificity of semantic fluency was found to be higher than MMSE and phonemic fluency in distinguishing individuals with ADD and MCI.Conclusion: The findings of the study show that MMSE is stronger than semantic and phonemic fluency in distinguishing healthy individuals, but its specificity is lower than semantic fluency in distinguishing MCI and AD dementia. It has become the most applied surgical procedure both in Turkiye, and globally. In 2013, Robson et al. created the Robson 10-Group Classification System (TGCS) in Ireland to evaluate and standardize CS indications more easily.Materials and MethodsThe data for this retrospective study were collected from the records of women, who gave birth between January 1, 2019 and July 31, 2022 at Mugla University Education and Research Hospital. These data were classified according to the TGCS.ResultsThere were 4889 deliveries in our clinic between the specified dates. 3177 (64.98%) of these births were CS. When the CS ratio was evaluated as a percentage, the groups with the highest ratio were, respectively, Robson groups 9, 5, 7, and 8 (100%, 99.34%, 97.56%, and 96.92%). When the CS ratio was evaluated as a percentage, the group with the lowest ratio was Robson group 3 (21.5%). When numerically evaluated in terms of the effect on the CS number, the ranking was Robson groups 5, 1, and 2, respectively (1,053, 893, and 568).ConclusionThe CS ratio of our clinic is above the mean for that of Turkiye, and the highest acceptable CS ratio according to the WHO. Since our clinic is the only university hospital in the city, all complicated pregnancies are referred to our clinic, with births also taking place there. These models in the framework of Structural Equation Modeling have been recommended as an alternative to classical methods such as analysis of variance. In this study, Latent Growth Models were introduced by using a Monte Carlo simulation approach and the interpretation of the findings was discussed. In addition, the effect of different sample sizes (30, 50, 100, and 200) on power and parameter estimates were examined. For this purpose; (1) data generation was performed with Monte Carlo simulation, (2) the parameters of unconditional and conditional models were estimated and the findings were discussed and (3) the effect of sample size on parameter estimates, standard errors, coverage and power was studied. All of the analyses were performed by using Mplus 5.1 software. This study aims to explore the relationship between corporate governance practices and financial sustainability in light of OECD principles of corporate governance. Altman's Z-scores and CG ratings of 20 companies listed in the Borsa Istanbul Corporate Governance Index (XKURY) were analyzed using the TOPSIS method. This study explores the hybrid nature of SME credit scoring systems by developing an SME credit scoring model with general indicators, financial indicators and intelligence indicators related to the company and the major shareholder. This study is the first to explore the effect of intelligence indicators in credit scoring. It used regression analysis to evaluate real data from 125 SMEs operating in Turkey. Of 17 independent variables, 9 had statistically significant relationships with SME credit scores. The paper includes a comprehensive discussion of the implications of these findings.
