Methods: Eighty-eight female patients with SjS and 3,338 age-matched healthy female controls were included in this study. 25 OH-D3 levels were compared with healthy controls. Then the patients were stratified according to their 25 OH-D3 levels, either insufficient/deficient or normal (<50 nmol/L or ≥50 nmol/L). The disease activity was evaluated using The EULAR SjS disease activity index (ESSDAI) and its components. Correlation analyses were also performed for a possible correlation with disease characteristics and markers of activity. Results: No differences in 25 OH-D3 levels were found between SjS and healthy populations (p>0.05). No correla- tions were found between patient characteristics or labo- ratory values (p>0.05). Conclusions: This study did not find a link between disease characteristics and disease activity and 25 OH-D3 levels. Although these vulnerabilities are often targeted individually, targeting all with an enzyme may prove a novel approach. However, therapeutic enzymes are prone to proteolytic degradation and neutralizing antibodies leading to a reduced half-life and effectiveness. We hypothesized that glucose oxidase (GOX) enzyme that catalyzes oxidation of glucose and production of hydrogen peroxide, may hit all these targets by depleting glucose; crippling anabolic pathways and producing reactive oxygen species (ROS); unbalancing redox homeostasis. Methods: We encapsulated GOX in an acrylamide layer and then performed activity assays in denaturizing settings to determine protection provided by encapsulation. Afterwards, we tested the effects of encapsulated (enGOX) and free (fGOX) enzyme on MCF-7 breast cancer cells. Results: GOX preserved 70% of its activity following encapsulation. When fGOX and enGOX treated with guanidinium chloride, fGOX lost approximately 72% of its activity, while enGOX only lost 30%. Both forms demonstrated remarkable resilience against degradation by proteinase K and inhibited viability of MCF-7 cells in an activity-dependent manner. Conclusions: Encapsulation provided protection to GOX against denaturation without reducing its activity, which would prolong half-life of the enzyme when administered intravenously.Article Citation - WoS: 2Citation - Scopus: 1Adjuvant Effects of Chemotherapeutics and Metformin on Mfe-319 Endometrial Carcinoma Cell Line(Editura Acad Romane, 2020) Aydemir, Isil; Uluer, Elgin Turkoz; Korkmaz, Oya; Tuglu, Mehmet Ibrahim; Inan, SevincWe aimed to investigate the cytotoxicity of Metformin, Cisplatin, and Paclitaxel on MFE-319 endometrial carcinoma cell line using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and immunocytochemistry assays. Half maximal inhibitory concentration (IC50) doses of three drugs alone and in the dual combinations were applied to the cells. Immunocytochemical method was performed for the cell survival and for phosphatidylinositol 3-kinase (PI3K), phosphorylated extracellular regulated kinases (pErk)-1/2, Akt-1, phosphorylated Akt (pAkt)-1/2/3 cell growth markers and angiogenic vascular endothelial growth factor (VEGF). Immunoreactivities were evaluated using H-score and analyzed using the one-way analysis of variance (ANOVA) test for statistics. It was found that these drugs caused a decrease in the immunoreactivities of these markers. Particularly, dual combination of Paclitaxel and Cisplatin decreased the immunoreactivities of PI3K, pErk-1/2, Akt-1, and pAkt-1/2/3. Cisplatin and Paclitaxel were more effective than Metformin; on the other hand, Metformin has been shown to enhance the efficacy of these two drugs. In vitro or in vivo further studies are needed to investigate the efficacy of these three drugs via PI3K/Akt signal pathway.Article Aerospace 3d Textile Preform Analysis: Experimental and Mesomodeling(Polish Society of Composite Materials, 2023) Saboktakin, Abbasali; Safaei, H.For the purpose of predicting how textile preforms affect the quality of the composite material and its performance, the stitched textile preform must be characterized. Experimental compaction analysis and finite element analysis of textile preforms are the main subjects of this paper. The formability parameters of a preform can be changed by the stitching process, according to research on the mechanical properties of preforms conducted during compression testing. The load-deformation response, which is depicted in detail, had the greatest influence on preform deformation. Less fiber bundle undulation in the plane direction and more stitching thread undulation in the thickness direction were observed during compression of the stitched preform, whereas the stitching thread improved the resistance of the preform to compression loading. © 2023, Polish Society of Composite Materials. Methods: The study adopted a retrospective design. It was carried out on 208 kidney transplant recipients. The social cognitive theory was used for understanding and addressing the issues of nonadherence to immunosuppressive medication. Data were collected with a self-reported-Immunosuppressant Therapy Adherence Scale and biological assays. Descriptive analyses and logistic regression were used to analyze data. Results: According to the self-reported assessment, medication adherence of the patients receiving transplants from live donors was 0.503 times lower than that of the patients receiving transplants from deceased donors. The biological assays did not show any affecting factors in medication adherence. Conclusion: The results of the study showed the importance of combining methods to assess medication adherence. In the self-reported assessment, live donor transplantation was found to negatively affect medication adherence. Turkiye is the country with the highest number of kidney transplantations from live donors in the world. Simple recipes and procedures were determined for the adaptation and use by anyone who is interested to use sustainable, non-toxic, and even edible plastic-like materials without the need for a scientific background. The recipes and procedures were developed by controlled laboratory experiments. The main recipes contain different combinations of agar strips, water, glycerine, and vinegar. A variety of films with different sensorial properties (soft, hard, flexible, stiff clear, coloured, textured, with or without spicy odour, etc.) were prepared by heating the ingredients in a saucepan, mixing, pouring on a flat surface, and drying for a few days. These were evaluated by tinkering and user studies that involved students from design, food engineering, culinary arts, and other fields. Yöntemler: Temmuz 2018 ile Ağustos 2019 tarihleri arasında aile hekimlerinden veri toplamak amacıyla çevrimiçi bir anket geliştirildi. Tanımlayıcı ve korelasyonel bir araştırma deseni kullanıldı. Çalışmada uygulanan anket, “katılımcıların kişisel bilgileri” ve “Sağlıklı Yaşam Biçimleri Davranış Ölçeği-II”i içermektedir. Kesitsel bir çalışma olarak tasarlanan bu araştırmada, örneklem kolayda örnekleme metodu ile sekiz ülkeden belirlenmiştir, ve anketler katılımcılara elektronik ortamda uygulanmıştır. Katılımcılara anketin süresi, veri saklama işlemleri, saklama süresi, araştırmacıların kimliği ve çalışmanın amacı hakkında bilgi verilmiştir. Bulgular: Çalışmaya 131 aile hekimi katılmış olup, erkek katılımcı oranı daha yüksek bulunmuştur. Sigara içme sıklığının Türk aile hekimleri arasında anlamlı derecede yüksek olduğu tespit edilmiştir (p=0,013). Ancak, ölçek toplam puanı veya altı boyutta alınan puanlar açısından aile hekimleri arasında istatistiksel olarak anlamlı bir fark bulunmamıştır. Sonuç: Çoğu aile hekimi, bu alanda daha fazla eğitim almak istediklerini belirtmiştir, bu da tıp fakültelerinin müfredatına eğitim müdahalelerin dahil edilmesi gerekliliğini göstermektedir. These models in the framework of Structural Equation Modeling have been recommended as an alternative to classical methods such as analysis of variance. In this study, Latent Growth Models were introduced by using a Monte Carlo simulation approach and the interpretation of the findings was discussed. In addition, the effect of different sample sizes (30, 50, 100, and 200) on power and parameter estimates were examined. For this purpose; (1) data generation was performed with Monte Carlo simulation, (2) the parameters of unconditional and conditional models were estimated and the findings were discussed and (3) the effect of sample size on parameter estimates, standard errors, coverage and power was studied. All of the analyses were performed by using Mplus 5.1 software. Therefore, the enterprises must give the necessary importance to consumers' desires and expectations. The consumers are affected by many factors such as quality, price and design while making a selection between different clothing brands. Design factor comes into prominence in recent years and color is one of the most significant factors of design. As it is already known, colors affect the buying behavior of consumers. This study aims to analyze the factors which affect the color preferences of consumers in clothing sector. In accordance with the aim of the research, a questionnaire form consisting of 20 questions is prepared. This survey is conducted within March 2014 at the hinterland of Izmir province residents by using face to face interview method. After the conduction of the survey, collected data are evaluated with SPSS 16.0 program. According to the results of our research, Turkish consumers give great importance to the colors and color harmony of garments which they have bought. Black, blue and navy blue are the most preferred garment colors whereas orange, pink and yellow are the least preferred ones. In addition to these, relationships between the demographic properties of consumers and color preferences are analyzed and sectoral suggestions are made.Article Citation - WoS: 2Citation - Scopus: 2Analysis of Nucleotide Changes in Rt-Pcr Primer/Probe Binding Regions in Sars-Cov Isolates Reported From Turkey(Ankara Microbiology Soc, 2021) Demir, Ayse Banu; Bulgurcu, Alihan; Appak, Ozgur; Sayiner, Ayca ArzuThe SARS-CoV-2 virus, which caused the COVID-19 epidemic, caused more than 55 million cases and nearly 1.5 million deaths worldwide. For the microbiological diagnosis of the disease, the most valid method is detecting the presence of the viral genome by real-time reverse transcription polymerase chain reaction (rRT-PCR). However, due to the nature of the RNA viruses, frequent mutations may affect the sensitivity of the analyses made on the genetic material of the virus, such as PCR. In this study, we aimed to investigate the mutations in the primer-probe binding regions of the rRT-PCR panels used in COVID-19 diagnosis. SARS-CoV-2 whole genome sequence data (n= 194) isolated from COVID-19 cases in Turkey and uploaded on GISAID database from the centers in Istanbul (n= 78), Ankara (n= 58), Kars (n= 47), Bursa (n= 2), Adiyaman (n= 2), Erciyes (n= 1) and Kocaeli (n= 1) between March 17-September 14, 2020 were analyzed. In order to determine the nucleotide changes, SARS-CoV-2 sequences from Turkey were compared to the reference genome sequence (NC_045512.1) present in GenBank website. The constructed data set was aligned using the MAFFT program and was checked manually if the sequences were in the same frame by using the AliView program. Primer-probe binding sites of the thirteen SARS-CoV-2 rRT-PCR panels from seven different institutes (US CDC, China CDC, Charite CDC, Pasteur, HKU, Thailand, NIID) that are being used in COVID-19 diagnosis were evaluated in terms of nucleotide changes within the corresponding regions compared to the reference genome. Sequence diversities in the viral genomes were determined via positional nucleotide numerical calculator and entropy calculator modules and nucleotide and entropy changes in primer-probe binding regions for each rRT-PCR panel were examined. Among thirteen different primer-probe panels, nucleotide changes in the target regions of the seven primer-probe panels were determined. When viral sequences with nucleotide changes in the primer-probe binding regions were examined, the most common changes were observed in the China CDC N-forward primer and US CDC N3-forward primer binding regions. It is important that the kits to be used as diagnostic tests are designed specific to the regions with less nucleotide changes. Nucleotide changes may not be critical for DNA amplification for most PCR panels, but should be carefully monitored as they may affect the sensitivity of the assay. If the risk of alteration of the designed region is high, the primer - probe binding sites should be checked frequently and updated when necessary.Article Citation - WoS: 10Citation - Scopus: 11An Analysis of the Sustainability Disclosures of Textile and Apparel Companies in Turkey(Ege Univ, 2019) Saygili, Ebru; Saygılı, Arıkan Tarık; Yargi, Seher GorenThe aim of this study is to discuss recent sustainability developments in the textile industry and to explore the disclosures of textile and apparel companies in Turkey. The economic, environmental and social sustainability disclosures of 34 companies were analyzed in relation to Global Reporting Initiative (GRI) standards. The results of the analysis revealed that the most frequently mentioned sustainability issues were related to employees, water, product, market, supplier, education, economic, energy and waste. A majority of companies provided economic disclosures about economic performance and anti-competition; environmental disclosures about water and effluents and energy; and social sustainability disclosures about training, education, employment, health and safety, diversity and equal opportunity, marketing and labeling.Review Antineutrophil Cytoplasmic Antibody-Associated Vasculitides: Clinical Manifestations and Pulmonary Involvement(Kare Publ, 2024) Parlak, E.Ş.; Şengül, A.; İn, E.Antineutrophil cytoplasmic antibody (ANCA)-associated vasculitides (AAV) are necrotizing autoimmune vasculitis resulting from immune-mediated damage to small and medium-sized vessels. Three primary clinicopathological syndromes are defined in AAV: Granulomatosis with polyangiitis (GPA), microscopic polyangiitis (MPA), and eosinophilic granulomatosis with polyangiitis (EGPA). The disease may present as alveolar hemorrhage due to a systemic inflammatory response, purpuric rash due to vascular rupture, or segmental glomerular infarction due to vascular occlusion. The lungs are commonly affected organs in AAV, with lung involvement categorized into five main groups: pulmonary capillaritis characterized by granulomatous inflammation (lung nodules), tracheobronchial inflammation, diffuse alveolar hemorrhage (DAH), interstitial lung disease (ILD), and asthma. DAH and ILD are associated with a poor prognosis. Materials and methods: Thirty-two healthy female Balb-c mice (6-8-week-old; weighing 22 +/- 5 g) were used in this study. The mice were randomly divided into four groups: control (n=8), BLM (n=8), BLM+EGCG (n=8), and EGCG (n=8). Skin tissue specimens were collected at the end of the experiments. Histopathological examinations of skin tissues were performed. Skin samples were assessed for total superoxide dismutase (SOD) activity and malondialdehyde (MDA) content. The phosphorylation of p-38 mitogen-activated protein kinase and Akt protein (the serine-threonine protein kinase encoded by the AKT), as well as the nuclear factor-kappa B (NF-kappa B) levels, were analyzed by western blotting. Results: Epigallocatechin-3-gallate-treated groups were observed to have reduced connective tissue fibrosis in the dermis area using Masson's trichrome staining method. Pp-38 and NF-kappa B were found to decrease significantly in the BLM + EGCG group compared with the BLM group. Parallel to these findings, phosphorylated Akt protein was found to increase in the BLM + EGCG group compared with the BLM group. SOD activity was increased in the EGCG group and content of MDA level was decreased in EGCG groups. WEDM erodes workpiece materials by a series of discrete electrical sparks between the workpiece and an electrode flushed or immersed in a dielectric fluid. The WEDM process is particularly suitable for machining hard materials as well as complex shapes. In this paper, a neural network and the Taguchi design method have been implemented for minimizing the surface roughness in a WEDM process. A back-propagation neural network (BPNN) was developed to predict the surface roughness. In the development of a predictive model, machining parameters of open-circuit voltage, pulse duration, wire speed and dielectric flushing pressure were considered as the input model variables of the AISI 4340 steel. An analysis of variance (ANOVA) was used to determine the significant parameter affecting the surface roughness (R-a). The objective functions have been selected in relation to parameters of FSW parameters; rotating speed, welding speed and tool shoulder diameter. The experiments were planned using Taguchi's L8 orthogonal array. Multi-response optimization was applied using Grey relation analysis and Taguchi approach to solve the problem. The significance of the factors on overall quality characteristics of the welding process has also been evaluated quantitatively by the analysis of variance (ANOVA) method. Optimal results have been verified through confirmation experiments. Data from these sensors allow the precise control of the laundry operation, often remotely. The combination of new machines with information technology has created a more efficient and cost effective process, enabled by software based on extensible architecture. The operations of a laundry exhibit all the properties of a context-aware system. This paper extends the authors' earlier work on context-aware laundry management architecture emphasizing business processes. In this study, we present architectural details that will aid implementers of the system in their choice of tools, techniques and technologies. In particular, a detailed account of laundry context information is presented. The proposed architectural framework used in the solution is able to monitor and autonomously manage laundry operations. The system employs a layered architecture which allows the separation of system components such as data capture, data processing and business services. An inference and a rule engine help to orchestrate system activities and transmit system status to interested parties through an interface. In this investigation method, an electromagnetic wave is transmitted using an antenna and the received signal is recorded. Detection of beam positions in this GPR data requires the skills of a trained human operator. This study utilized a multi-layer neural network to detect beam positions in the GPR data. The visual description and definition of GPR data has major disadvantages and a neural network has been studied to overcome these shortcomings. A set of 32,740 training vectors with a length of 64 data was implemented to train the neural network. A new set of 16,370 testing vectors with a length of 64 data was then prepared to test the performance. In order to create something new, a move away from conventional generic shapes toward more interesting structures is a necessity. The purpose of this study was to investigate an approach to the design process that could facilitate the understanding of structure as a fundamental component of fashion design and support students' development in creativity with structurally interesting garments. By developing a concept based on nature using 2-dimensional visual source material, drawing, collage, and experimental research development in the form of a sketchbook, a group of students from Department of Fashion Design who joined the project, were encouraged to make a rational analysis of the structural elements at their disposal. By executing a series of exercises aimed at exploring structure, students were observed to make a gradual progression from 2-dimensional research through design development, to 3-dimensional garment construction. In conjunction with the idea of structurally interesting designs, fabric choices, pattern making and garment construction were also considered. Each step from 2-dimensional exercises to garment construction was recorded as visual and written documentation. Additionally, a questionnaire was developed to receive student opinions and feedback. As a result, configuration of the design exercises and the project setup has been identified as an encouraging approach both to develop students' understanding of structure in fashion design and for the development of creativity in 3-dimension.
