Abstract
Many treatment strategies and efforts have been made to develop effective tumor targeting drugs that cause minimal toxicity in body, including radiopharmaceutical therapy and radioimmunotherapy. However, selection of linker molecules between the radioactive metals and biological drugs are critical to design stable radiopharmaceuticals. In this study, conjugation of trastuzumab ( humanized monoclonal antibody) with different chelating molecules (dodecane tetraacetic acid (DOTA), ethylenediaminetetraacetic acid (EDTA) and diethylene triamine pentaacetic acid (DTPA)) has been investigated by using molecular docking and spectroscopic methods (UV-Vis, Fluorescence). We found that the trastuzumab+DOTA complex exhibits a more stable binding mechanism together with minute structural changes.
