Article 1,25-Dihydroxyvitamin D3 Induces N-Myc Downstream Regulated Gene-2 Expression in Papillary Thyroid Carcinoma Cells(Dokuz Eylul Univ Inst Health Sciences, 2020) Sipahi, Murat; Bartik, Didem Keles; Doruk, Mehmet; Bayraktar, Firat; Oktay, GulgunPurpose: In addition to its role in serum calcium homeostasis, the anti-tumor function of 1,25-dihydroxyvitamin D-3 (calcitriol) in cancer development is well established. N-myc Downstream Regulated Gene 2 which functions as a tumor suppressor gene has recently been shown to be downregulated in various cancer leading to increased tumor incidence, progression and metastasis. The goal of this study was to investigate the possible effects of calcitriol treatment on NDRG2 expression in BCPAP papillary thyroid carcinoma cells. Methods: The experiments were carried on human primary thyroid follicular epithelial cells (Nthy-ori-3-1), and human papillary thyroid carcinoma cells (BCPAP). The half maximal inhibitory concentration (IC 50) of calcitriol on BCPAP cells was determined by WST-1 assay. BCPAP cells were treated with 15 and 30 mu M calcitriol for 24, 48, and 72 hours, respectively. Basal NDGR2 expression in Nthy-ori-3-1 and BCPAP cells as well as the alterations on NDRG2 expression in calcitriol treated BCPAP cells were evaluated with western blot. Results: A significant downregulation of NDRG2 was observed in BCPAP cells when compared to Nthy-ori-3-1 cells (p<0.01). IC50 dose of calcitriol was found to be 64, 54 and 43 mu M for 24, 48 and 72 hours, respectively. NDRG2 protein expression levels were significantly increased in 30 mu M calcitriol treated BCPAP cells after 48 hours (p<0.05). Conclusions: Calcitriol induced NDRG2 protein expression in BCPAP cells. We predict that calcitriol increased NDRG2 protein levels in BCPAP cells via c-Myc repression, which is upregulated by aberrant Wnt/beta-catenin signaling. Methods: Eighty-eight female patients with SjS and 3,338 age-matched healthy female controls were included in this study. 25 OH-D3 levels were compared with healthy controls. Then the patients were stratified according to their 25 OH-D3 levels, either insufficient/deficient or normal (<50 nmol/L or ≥50 nmol/L). The disease activity was evaluated using The EULAR SjS disease activity index (ESSDAI) and its components. Correlation analyses were also performed for a possible correlation with disease characteristics and markers of activity. Results: No differences in 25 OH-D3 levels were found between SjS and healthy populations (p>0.05). No correla- tions were found between patient characteristics or labo- ratory values (p>0.05). Conclusions: This study did not find a link between disease characteristics and disease activity and 25 OH-D3 levels. MATERIALS AND METHODS: This study included 122 people and was administered in a smoking cessation clinic. The sociodemographic-smoking status characteristics questionnaire and the ARME scale were used for data collection. A psycholinguistic language adaptation was performed. In the validity, analyses, content, construct, and criterion-related validities were used. For content validity, expert evaluation was performed. For construct validity, principal component analyses (exploratory factor analyses) were performed. Orthogonal (Varimax) rotation was used to explore multiple factors. The Kaiser-Meyer-Olkin test was used to assess the adequacy of the sample size. For criterion-related validity, we compared the ARME scale points of people who were abstinent and had relapse for smoking at the end of the sixth month. In the reliability analysis, standard deviation (SD) and item analysis, internal consistency, and test-retest methods were used. RESULTS: The four factors explain 58% of the total variance. Items have factor loading between 0.409 and 0.805. When the factor structure of the scale was assessed, the items in each factor group have a factor load of at least 0.40. Due to one-dimensional use of the original scale, it has been decided to maintain this scale in its original form. The ARME scale points of people who quit smoking were statistically higher than the points of people who had relapse at the end of the sixth month. Cronbach's alpha coefficients were between 0.846 and 0.763. Significant and positive correlation was found between the test-retest scale scores. CONCLUSION: The Turkish adaptation of the ARME scale, which was developed for adults who quitted smoking, is an adequately valid and reliable measurement instrument. It is considered that the scale might be used reliably in different cultures as well.Article Citation - WoS: 2Accidental Humor in Consecutive Interpreting: Accidentally Useful Food for Thought(Istanbul Univ, Fac Letters, 2021) Yetkin Karakoç, NihalUnlike humor translation, humor in translation, caused by translation errors, is underresearched in translation studies. Accidental humor, as a subgroup of humor, has been analyzed in translation contexts, but to the best knowledge of the author, not in the interpreting context. This descriptive study, being the first of its kind dealing with the accidental humor in consecutive interpreting, is intended to fill the gap in the verbal manifestations of resources in the English-Turkish interpreting context along with the possible reasons for resources of accidental humor in consecutive interpreting class. As a result of the study, a total of 38 accidentally humorous instances are found. The verbal resources are analyzed and divided into main and sub-categories, and for each, examples from the corpus are discussed. In addition, new linguistic resources were discovered, which may be exclusive to this context. The detected items are by no means meant to be exhaustive, as also stated by the former studies, but nevertheless enables greater understanding about the types and possible causes of accidental humor which may occur during the consecutive interpreting process. Given the dearth of literature on accidental humor in consecutive interpreting, this study may provide some insight for remedial teaching/learning in interpreting studies, as well as linguistics. Future studies may examine, among the others, verbal resources in the simultaneous interpreting context and compare the verbal resources in depth in consecutive with those in simultaneous interpreting.Article Aconitine Impedes Cell Motility in Mda-Mb Breast Cancer Cells(Dokuz Eylul Univ Inst Health Sciences, 2024) Keles, Didem; Sipahi, Murat; Surer, Seniz Inanc; Oktay, GulgunPurpose: Aconitine, a potent alkaloid from Aconitum plants, has shown promising anticancer properties. The aim of the study is to investigate the effects of aconitine on lateral migration, and matrix metalloproteinase (MMP) activity in MDA-MB-231 triple-negative breast cancer cells. Material and Methods: A WST-1 viability assay was conducted to determine the effect of aconitine on the viability of MDA-MB-231 cells. Following treatment with non-cytotoxic doses of aconitine, lateral migration was evaluated through wound healing assays. Additionally, gelatin zymography was conducted to analyze MMP-2 and MMP-9 activity and secretion levels. Results: Aconitine concentrations up to 200 mu M did not significantly affect cell viability for up to 72 hours, whereas higher doses (400-600 mu M) reduced viability in a time-dependent manner. Aconitine at 200 mu M showed a trend towards decreased lateral motility, with a significant reduction at 9 hours post-treatment. Gelatin zymography revealed no alterations in MMP-2 and MMP-9 activity or secretion levels following aconitine treatment. Conclusion: Aconitine demonstrates limited efficacy in modulating the migratory capacity of MDA-MB231 cells and does not affect gelatinase activity. Although these vulnerabilities are often targeted individually, targeting all with an enzyme may prove a novel approach. However, therapeutic enzymes are prone to proteolytic degradation and neutralizing antibodies leading to a reduced half-life and effectiveness. We hypothesized that glucose oxidase (GOX) enzyme that catalyzes oxidation of glucose and production of hydrogen peroxide, may hit all these targets by depleting glucose; crippling anabolic pathways and producing reactive oxygen species (ROS); unbalancing redox homeostasis. Methods: We encapsulated GOX in an acrylamide layer and then performed activity assays in denaturizing settings to determine protection provided by encapsulation. Afterwards, we tested the effects of encapsulated (enGOX) and free (fGOX) enzyme on MCF-7 breast cancer cells. Results: GOX preserved 70% of its activity following encapsulation. When fGOX and enGOX treated with guanidinium chloride, fGOX lost approximately 72% of its activity, while enGOX only lost 30%. Both forms demonstrated remarkable resilience against degradation by proteinase K and inhibited viability of MCF-7 cells in an activity-dependent manner. Conclusions: Encapsulation provided protection to GOX against denaturation without reducing its activity, which would prolong half-life of the enzyme when administered intravenously.Article Citation - WoS: 2Citation - Scopus: 2Adrenomedullin Has No Effect on Segmental Bone Defect Healing but Increases Bone Mineral Density in Rat Model(AVES, 2023) Kaymakoğlu, Mehmet; Ciftci, E.; Korkusuz, P.; Ozdemir, E.; Erden, M.E.; Turhan, E.Objective: This study aimed to investigate the effect of adrenomedullin on the healing of the segmental bone defect in a rat model. Methods: Thirty-six Wistar rats were randomly divided into 6 groups based on follow-up periods and administered a dose of adreno-medullin hormone. In each group, bilaterally, a 2-mm bone defect was created at the diaphysis of the radius. Sodium chloride solution was administered to sham groups 3 times a week for 4 and 8 weeks intraperitoneally. Adrenomedullin was administered to the study groups 3 times a week: 15 μg—4 weeks, 15 μg—8 weeks, 30 μg—4 weeks, and 30 μg—8 weeks, respectively. After euthanasia, the segmental defects were evaluated by histomorphometric [new bone area (NBA)] and microtomographic [bone volume (BV), bone surface (BS), and bone mineral density (BMD)] analyses. Results: Although the 4-and 8-week 15 μg administered study groups had higher NBA values than the other study and control groups, the histomorphometric analysis did not reveal any statistical difference between the control and study groups regarding NBA (P >.05). In microtomographic analysis, BV was higher in the 15 μg 4-week group than 30 μg 4-week group (296.9 vs. 208.5, P =.003), and BS was lower in the 30 μg 4-week group than in the 4-week control group (695.5 vs. 1334.7, P =.005), but overall, no significant difference was found between the control and study groups (P >.05). Despite these minor differences in histomorphometric and microtomographic criteria indicating new bone formation, the BMD values of the 15 μg 8-week study group showed a significant increase compared with the control group (P =.001, respectively). Conclusion: Adrenomedullin positively affected BMD at 15 μg, but this study could not show healing in the segmental defect site at different dose regimens. Further studies are needed to assess its effects on bone tissue trauma. © 2023, AVES. Despite many controversies about how Karagoz came into existence, there is a consensus that it emerged in the 16th century. Figures, as cultural objects, have production methods and techniques interwoven with the doctrines of the past. Today, figure-making for different purposes, inherited from the past, is present in Karagoz figure art. The aesthetic reflections, technique, and material harmony of the figures for show purposes must carry the cultural essence of Karagoz. This study examines museum collections and exhibitions, interviews with Karagoz masters, and a data analysis from an academic perspective. Considering these factors, it becomes clear that the training of masters for figure-making art is significant. Long-term training organized by institutions will contribute to this art and might be beneficial for good examples to reach the audience. Methods: The study adopted a retrospective design. It was carried out on 208 kidney transplant recipients. The social cognitive theory was used for understanding and addressing the issues of nonadherence to immunosuppressive medication. Data were collected with a self-reported-Immunosuppressant Therapy Adherence Scale and biological assays. Descriptive analyses and logistic regression were used to analyze data. Results: According to the self-reported assessment, medication adherence of the patients receiving transplants from live donors was 0.503 times lower than that of the patients receiving transplants from deceased donors. The biological assays did not show any affecting factors in medication adherence. Conclusion: The results of the study showed the importance of combining methods to assess medication adherence. In the self-reported assessment, live donor transplantation was found to negatively affect medication adherence. Turkiye is the country with the highest number of kidney transplantations from live donors in the world. As a proactive approach, lifelong learning presents an opportunity for the elderly to challenge stereotypes, remain engaged in society, and foster personal growth. Yöntemler: Temmuz 2018 ile Ağustos 2019 tarihleri arasında aile hekimlerinden veri toplamak amacıyla çevrimiçi bir anket geliştirildi. Tanımlayıcı ve korelasyonel bir araştırma deseni kullanıldı. Çalışmada uygulanan anket, “katılımcıların kişisel bilgileri” ve “Sağlıklı Yaşam Biçimleri Davranış Ölçeği-II”i içermektedir. Kesitsel bir çalışma olarak tasarlanan bu araştırmada, örneklem kolayda örnekleme metodu ile sekiz ülkeden belirlenmiştir, ve anketler katılımcılara elektronik ortamda uygulanmıştır. Katılımcılara anketin süresi, veri saklama işlemleri, saklama süresi, araştırmacıların kimliği ve çalışmanın amacı hakkında bilgi verilmiştir. Bulgular: Çalışmaya 131 aile hekimi katılmış olup, erkek katılımcı oranı daha yüksek bulunmuştur. Sigara içme sıklığının Türk aile hekimleri arasında anlamlı derecede yüksek olduğu tespit edilmiştir (p=0,013). Ancak, ölçek toplam puanı veya altı boyutta alınan puanlar açısından aile hekimleri arasında istatistiksel olarak anlamlı bir fark bulunmamıştır. Sonuç: Çoğu aile hekimi, bu alanda daha fazla eğitim almak istediklerini belirtmiştir, bu da tıp fakültelerinin müfredatına eğitim müdahalelerin dahil edilmesi gerekliliğini göstermektedir. Ayrıca, iyi yapılandırılmış web tabanlı kitlesel çevrimiçi kurslar, aile hekimlerinin sağlıklı yaşam biçimi davranışları ve yaşam biçimi tıbbı uygulamalarına yönelik eğitim gereksinimlerini karşılamada etkili bir çözüm olabilir.Article Alternative E-Assessment in Language Classes: Recent Experiences and Future Directions(Ani Publishing, 2024) Çağlayan, EsinThis study aimed to explore English as a Foreign Language (EFL) teachers' perceptions regarding alternative e-assessments in the Turkish tertiary level context. The study utilized semi-structured interviews with a sample of EFL instructors from 17 higher education institutions in Türkiye. The data were analyzed using thematic analysis to identify emerging themes. The results revealed varying perceptions about alternative e-assessments, with six primary themes emerging: impact on learning, workload, academic integrity, digital technology, grading and feedback, and assessment competence. Overall, this qualitative study provides valuable insights into the perceptions of EFL teachers regarding alternative e-assessments, highlighting key areas of concern. The study emphasizes the need to address these concerns in professional development programs that provide teachers with the necessary knowledge and skills to effectively implement alternative e-assessments. The results also have implications for the development of effective assessment policies and procedures, optimizing educational practices in the digital age.Article Alzheimer Hastaliği Klinik Olcutlerine Gore Demans ve Hafif Kognitif Bozukluk Ayriminda Sozel Akicilik Testlerinin Mini Mental Durum Testi ile Birlikte Kullanilmasi Yararli Olabilir(Cukurova Univ, Fac Medicine, 2022) Isbitiren, Yagmur Ozbek; Kiyi, Ilayda; Yener, GörsevPurpose: The aim of this study is to present the sensitivity, specificity and cut-off points of Mini Mental State Examination (MMSE), semantic and phonemic fluency tests determined by ROC curve analysis in differentiating between Alzheimer's dementia (AD) and mild cognitive impairment (MCI).Materials and Methods: Age-education-sex matched 72 healthy individuals, 72 MCI and 72 AD dementia individuals were included in the study. MMSE, semantic and phonemic fluency tests were applied to all participants. Sensitivity and specificity values and cut-off scores of MMSE, semantic and phonemic fluency tests in distinguishing healthy, MCI and ADD individuals were obtained by ROC curve analysis.Results: ROC analyses show that all three tests can significantly discriminate between healthy, ADD and MCI individuals. It was seen that the discrimination power of the MMSE was higher than the verbal fluency tests when the areas under the ROC curve were compared. The specificity of semantic fluency was found to be higher than MMSE and phonemic fluency in distinguishing individuals with ADD and MCI.Conclusion: The findings of the study show that MMSE is stronger than semantic and phonemic fluency in distinguishing healthy individuals, but its specificity is lower than semantic fluency in distinguishing MCI and AD dementia. This study explores the hybrid nature of SME credit scoring systems by developing an SME credit scoring model with general indicators, financial indicators and intelligence indicators related to the company and the major shareholder. This study is the first to explore the effect of intelligence indicators in credit scoring. It used regression analysis to evaluate real data from 125 SMEs operating in Turkey. Of 17 independent variables, 9 had statistically significant relationships with SME credit scores. For the microbiological diagnosis of the disease, the most valid method is detecting the presence of the viral genome by real-time reverse transcription polymerase chain reaction (rRT-PCR). However, due to the nature of the RNA viruses, frequent mutations may affect the sensitivity of the analyses made on the genetic material of the virus, such as PCR. In this study, we aimed to investigate the mutations in the primer-probe binding regions of the rRT-PCR panels used in COVID-19 diagnosis. SARS-CoV-2 whole genome sequence data (n= 194) isolated from COVID-19 cases in Turkey and uploaded on GISAID database from the centers in Istanbul (n= 78), Ankara (n= 58), Kars (n= 47), Bursa (n= 2), Adiyaman (n= 2), Erciyes (n= 1) and Kocaeli (n= 1) between March 17-September 14, 2020 were analyzed. In order to determine the nucleotide changes, SARS-CoV-2 sequences from Turkey were compared to the reference genome sequence (NC_045512.1) present in GenBank website. The constructed data set was aligned using the MAFFT program and was checked manually if the sequences were in the same frame by using the AliView program. Primer-probe binding sites of the thirteen SARS-CoV-2 rRT-PCR panels from seven different institutes (US CDC, China CDC, Charite CDC, Pasteur, HKU, Thailand, NIID) that are being used in COVID-19 diagnosis were evaluated in terms of nucleotide changes within the corresponding regions compared to the reference genome. Sequence diversities in the viral genomes were determined via positional nucleotide numerical calculator and entropy calculator modules and nucleotide and entropy changes in primer-probe binding regions for each rRT-PCR panel were examined. Among thirteen different primer-probe panels, nucleotide changes in the target regions of the seven primer-probe panels were determined. When viral sequences with nucleotide changes in the primer-probe binding regions were examined, the most common changes were observed in the China CDC N-forward primer and US CDC N3-forward primer binding regions. It is important that the kits to be used as diagnostic tests are designed specific to the regions with less nucleotide changes. Nucleotide changes may not be critical for DNA amplification for most PCR panels, but should be carefully monitored as they may affect the sensitivity of the assay. Interviews were conducted with earthquake victims in the city of Kocaeli in Turkey, and needs were identified. Also, user survey results from previous studies that have been conducted after the August 17th earthquake in 1999 were used to establish the design requirements for a temporary housing unit. While lack of space and issues of crowding, noise, and hygiene were mentioned as the most important problems in temporary earthquake shelters, suggestions to improve them included consideration of psychology and privacy needs of victims. Methods: In this retrospective study, a total of 314 patients who were operated on for hip fractures were included study. Patients were then divided into four groups based in their implant types: long-stem cementless bipolar hemiarthroplasty (n = 124; 102 female, 22 male; mean age = 84.2 +/- 6.4 years), standard-stem cementless bipolar hemiarthroplasty (n = 74; 48 female, 26 male; mean age = 83.5 +/- 6.9 years), antegrade intertrochanteric nail (n = 61; 35 female, 26 male; mean age = 78.5 +/- 6.8 years), and total hip arthroplasty (n = 55; 34 female, 21 male; mean age = 72.5 +/- 4.3 years). Data including gender, age, duration from injury to surgery, American Society of Anesthesiologists (ASA) score, comorbidities, use of antiplatelet agents, Barthel Index of Activities of Daily Living, type of anesthesia, operation time, preoperative hemoglobin values, blood transfusions given, duration of hospital stay, complications, and type of fracture were recorded. Results: Overall, the mean follow-up was 36.5 (range = 0 - 107) months. The overall mortality rate was 53.2%. The median survival duration was 44.2 +/- 5 months (range = 34.3 - 54). Survival rates were found significantly different among the groups (P = 0.001). In the first three years postoperatively, the mortality rate was higher in the standard-stem bipolar hemiarthroplasty group, but in the long-term follow-up, the long-stem bipolar hemiarthroplasty group exhibited the higher mortality rates. It was observed that some parameters had statistically significant effects on the mortality rates. Male gender, higher age, lower hemoglobin values, increased number of blood transfusions, ASA scores >= 3, the existence of >= 3 comorbidities were found as main predictors of increased mortality rates. Conclusion: The results of this study have shown that age, gender, preoperative hemoglobin levels, ASA scores, and comorbidities are significant factors affecting mortality in elderly patients operated on for hip fractures. Long-stem cementless bipolar hemiarthroplasty appears to show similar rates of mortality with standard-stem cementless bipolar hemiarthroplasty.Article An Anthropomorphistic Approach To Measuring Civil Society Organization's Reputation: a Semantic Network Analysis(Marmara Univ, Fac Communication, 2018) Turkel, SelinThe aim of the study is to reveal common meaning of reputation and disreputableness for civil society organizations (CSOs) by utilizing anthropomorphism. Accordingly, 212 individuals living in Izmir were asked to personify the CSOs they deemed reputable and disreputable, and provide adjectives that best described CSOs. In this study which was designed as descriptive research, semantic network analysis was employed and reputable and disreputable networks were visualized using a social network analysis software named Pajek. The relational analysis was performed using the measures of m-core, clique, articulation point, in-out degree, in-out closeness and betweenness. The first study to uncover the shared meaning of the CSO's reputation adopting the anthropomorphism approach reveals both positive and negative personality traits such as being helpful, honest, reliable and self-seeking, trickster, liar, and untrustworthy. The results of the research can contribute to the development of an appropriate measure of CSO's reputation for Turkish society.
