Article 1,25-Dihydroxyvitamin D3 Induces N-Myc Downstream Regulated Gene-2 Expression in Papillary Thyroid Carcinoma Cells(Dokuz Eylul Univ Inst Health Sciences, 2020) Sipahi, Murat; Bartik, Didem Keles; Doruk, Mehmet; Bayraktar, Firat; Oktay, GulgunPurpose: In addition to its role in serum calcium homeostasis, the anti-tumor function of 1,25-dihydroxyvitamin D-3 (calcitriol) in cancer development is well established. N-myc Downstream Regulated Gene 2 which functions as a tumor suppressor gene has recently been shown to be downregulated in various cancer leading to increased tumor incidence, progression and metastasis. The goal of this study was to investigate the possible effects of calcitriol treatment on NDRG2 expression in BCPAP papillary thyroid carcinoma cells. Methods: The experiments were carried on human primary thyroid follicular epithelial cells (Nthy-ori-3-1), and human papillary thyroid carcinoma cells (BCPAP). The half maximal inhibitory concentration (IC 50) of calcitriol on BCPAP cells was determined by WST-1 assay. BCPAP cells were treated with 15 and 30 mu M calcitriol for 24, 48, and 72 hours, respectively. Basal NDGR2 expression in Nthy-ori-3-1 and BCPAP cells as well as the alterations on NDRG2 expression in calcitriol treated BCPAP cells were evaluated with western blot. Results: A significant downregulation of NDRG2 was observed in BCPAP cells when compared to Nthy-ori-3-1 cells (p<0.01). IC50 dose of calcitriol was found to be 64, 54 and 43 mu M for 24, 48 and 72 hours, respectively. NDRG2 protein expression levels were significantly increased in 30 mu M calcitriol treated BCPAP cells after 48 hours (p<0.05). Conclusions: Calcitriol induced NDRG2 protein expression in BCPAP cells. We predict that calcitriol increased NDRG2 protein levels in BCPAP cells via c-Myc repression, which is upregulated by aberrant Wnt/beta-catenin signaling. Further investigation is required to enlighten the possible effect mechanisms of calcitriol in BCPAP cells.Article 16. Yüzyıl Giysi Tarihi Yazımı Üzerine: Giysilerde Doğu-batı Etkileşimi, Egzotizm ve Güç(2013) Er, Fehmiye DilekModa tarihinde batılı-olmayan giysilerin sunum ve dekorasyonları her zaman fantastik, büyüleyici, epik, dikkat çekici, egzotik, ruhani, zamansız ve değişmez gibi sözlerle ifade edilmiştir. Doğulu giysiler ve görünümler üzerindeki bu yaklaşım batılı anlayışın diğer kültürler üzerindeki yüceliğini, farklılığını ve gücünü gösterme arzusuyla yakından ilişkilidir. 19. yüzyıldan başlamak suretiyle doğunun dikkat çekici sunumları, batı giysi tarihinde batılı olmayan giysilerdeki teknik ustalıklar ve bu giysilerdeki özgün motifler egzotik olarak değerlendirilmiştir. Bu amaçla hem giysi tarihi hem de modern moda tarihi yazımında doğunun her zaman egzotik, primitif, öteki ve otantik olma noktasında bir söylem söz konusu olmuştur. Özellikle 16. yüzyıl Osmanlı İmparatorluğu?na ait egzotik giysiler ve kumaş teknikleri aynı dönem Avrupa modasına yoğun bir şekilde adapte edilirken giysiler ihtişamın ve gücün de bir göstergesi olmuşlardır. Bu çalışmada giysi tarihi açısından Osmanlı İmparatorluğu?nun en gösterişli dönemlerinden biri olan 16. yüzyılda giyilen Avrupalı giysilerinde Osmanlı giysi kültürünün izleri, egzotizm kavramı çerçevesinden incelenecektir. Bu çalışmada batılı giysilerdeki egzotik gösterimler aracılığı ile okumalar yapılırken, “egzotizm” kavramı, batıya ait olmayanı anlatma noktasında ele alınacaktır. How Turkish media influenced the constitution making process including workings of Constitutional Conciliation Commission is discussed, surveying relevant media content or performance with a particular focus on the political economy of Turkish media during that period. Traditional media outlets such as newspapers and television channels, and nowadays largely digital platforms, function as present-day agoras where political agenda is determined and contested. These online and offline channels have different levels of influence on discussions of the political elite, as presented extensively in media and communication studies. Together with the global media environment, national media foster the emergence of its own elites, while maintaining multi-layered relationships with high-level political decision-making processes. Media channels therefore bear symbolic powers that impact political policymaking by employing methods such as agenda setting and framing. Methods: Eighty-eight female patients with SjS and 3,338 age-matched healthy female controls were included in this study. 25 OH-D3 levels were compared with healthy controls. Then the patients were stratified according to their 25 OH-D3 levels, either insufficient/deficient or normal (<50 nmol/L or ≥50 nmol/L). The disease activity was evaluated using The EULAR SjS disease activity index (ESSDAI) and its components. Correlation analyses were also performed for a possible correlation with disease characteristics and markers of activity. Results: No differences in 25 OH-D3 levels were found between SjS and healthy populations (p>0.05). No correla- tions were found between patient characteristics or labo- ratory values (p>0.05). Conclusions: This study did not find a link between disease characteristics and disease activity and 25 OH-D3 levels. İzin vermeye yetkili makam anlamına gelen yetkili merci kararlarının yine “idari” nitelikte olacağı öngörülen itiraz merci kararıyla inceleneceği öngörülmüştür. Deneyler sonucunda en ideal yüzey kalitesinin elde edildiği kesme hızı, ilerleme miktarı, talaş derinliği ve kesici uç belirlenmiştir. Sonuç olarak, iki farklı mekanik dayanıma sahip alüminyum alaşımının yüzey sonuçları karşılaştırılıp optimum işleme değerleri bulunmuştur. The vertebral arteries running towards cranial cavity aresurrounded and protected by foramina transversarium of upper six cervical vertebrae. Surgeons should be aware of this variation beforeoperations to neck region, because the vertebral arteries may easily be injured due to lack of anterior bony protection of the costalelements. MATERIALS AND METHODS: This study included 122 people and was administered in a smoking cessation clinic. The sociodemographic-smoking status characteristics questionnaire and the ARME scale were used for data collection. A psycholinguistic language adaptation was performed. In the validity, analyses, content, construct, and criterion-related validities were used. For content validity, expert evaluation was performed. For construct validity, principal component analyses (exploratory factor analyses) were performed. Orthogonal (Varimax) rotation was used to explore multiple factors. The Kaiser-Meyer-Olkin test was used to assess the adequacy of the sample size. For criterion-related validity, we compared the ARME scale points of people who were abstinent and had relapse for smoking at the end of the sixth month. In the reliability analysis, standard deviation (SD) and item analysis, internal consistency, and test-retest methods were used. RESULTS: The four factors explain 58% of the total variance. Items have factor loading between 0.409 and 0.805. When the factor structure of the scale was assessed, the items in each factor group have a factor load of at least 0.40. Due to one-dimensional use of the original scale, it has been decided to maintain this scale in its original form. The ARME scale points of people who quit smoking were statistically higher than the points of people who had relapse at the end of the sixth month. Cronbach's alpha coefficients were between 0.846 and 0.763. Significant and positive correlation was found between the test-retest scale scores. CONCLUSION: The Turkish adaptation of the ARME scale, which was developed for adults who quitted smoking, is an adequately valid and reliable measurement instrument. It is considered that the scale might be used reliably in different cultures as well.Article Citation - WoS: 2Accidental Humor in Consecutive Interpreting: Accidentally Useful Food for Thought(Istanbul Univ, Fac Letters, 2021) Yetkin Karakoç, NihalUnlike humor translation, humor in translation, caused by translation errors, is underresearched in translation studies. Accidental humor, as a subgroup of humor, has been analyzed in translation contexts, but to the best knowledge of the author, not in the interpreting context. This descriptive study, being the first of its kind dealing with the accidental humor in consecutive interpreting, is intended to fill the gap in the verbal manifestations of resources in the English-Turkish interpreting context along with the possible reasons for resources of accidental humor in consecutive interpreting class. As a result of the study, a total of 38 accidentally humorous instances are found. The verbal resources are analyzed and divided into main and sub-categories, and for each, examples from the corpus are discussed. In addition, new linguistic resources were discovered, which may be exclusive to this context. The detected items are by no means meant to be exhaustive, as also stated by the former studies, but nevertheless enables greater understanding about the types and possible causes of accidental humor which may occur during the consecutive interpreting process. Given the dearth of literature on accidental humor in consecutive interpreting, this study may provide some insight for remedial teaching/learning in interpreting studies, as well as linguistics. Future studies may examine, among the others, verbal resources in the simultaneous interpreting context and compare the verbal resources in depth in consecutive with those in simultaneous interpreting.Article Aconitine Impedes Cell Motility in Mda-Mb Breast Cancer Cells(Dokuz Eylul Univ Inst Health Sciences, 2024) Keles, Didem; Sipahi, Murat; Surer, Seniz Inanc; Oktay, GulgunPurpose: Aconitine, a potent alkaloid from Aconitum plants, has shown promising anticancer properties. The aim of the study is to investigate the effects of aconitine on lateral migration, and matrix metalloproteinase (MMP) activity in MDA-MB-231 triple-negative breast cancer cells. Material and Methods: A WST-1 viability assay was conducted to determine the effect of aconitine on the viability of MDA-MB-231 cells. Following treatment with non-cytotoxic doses of aconitine, lateral migration was evaluated through wound healing assays. Additionally, gelatin zymography was conducted to analyze MMP-2 and MMP-9 activity and secretion levels. Results: Aconitine concentrations up to 200 mu M did not significantly affect cell viability for up to 72 hours, whereas higher doses (400-600 mu M) reduced viability in a time-dependent manner. Aconitine at 200 mu M showed a trend towards decreased lateral motility, with a significant reduction at 9 hours post-treatment. Gelatin zymography revealed no alterations in MMP-2 and MMP-9 activity or secretion levels following aconitine treatment. Conclusion: Aconitine demonstrates limited efficacy in modulating the migratory capacity of MDA-MB231 cells and does not affect gelatinase activity. Further investigation into underlying mechanisms is necessary, potentially leading to novel therapeutic strategies for triple-negative breast cancer.Article Citation - WoS: 6Citation - Scopus: 5Acrylamide-Encapsulated Glucose Oxidase Inhibits Breast Cancer Cell Viability(Walter De Gruyter Gmbh, 2020) Rrustemi, Trendelina; Geyik, Oyku Gonul; Ozkaya, Ali Burak; Ozturk, Taylan Kurtulus; Yuce, Zeynep; Kilinc, AliObjectives: Cancer cells modulate metabolic pathways to ensure continuity of energy, macromolecules and redoxhomeostasis. Although these vulnerabilities are often targeted individually, targeting all with an enzyme may prove a novel approach. However, therapeutic enzymes are prone to proteolytic degradation and neutralizing antibodies leading to a reduced half-life and effectiveness. We hypothesized that glucose oxidase (GOX) enzyme that catalyzes oxidation of glucose and production of hydrogen peroxide, may hit all these targets by depleting glucose; crippling anabolic pathways and producing reactive oxygen species (ROS); unbalancing redox homeostasis. Methods: We encapsulated GOX in an acrylamide layer and then performed activity assays in denaturizing settings to determine protection provided by encapsulation. Afterwards, we tested the effects of encapsulated (enGOX) and free (fGOX) enzyme on MCF-7 breast cancer cells. Results: GOX preserved 70% of its activity following encapsulation. When fGOX and enGOX treated with guanidinium chloride, fGOX lost approximately 72% of its activity, while enGOX only lost 30%. Both forms demonstrated remarkable resilience against degradation by proteinase K and inhibited viability of MCF-7 cells in an activity-dependent manner. Conclusions: Encapsulation provided protection to GOX against denaturation without reducing its activity, which would prolong half-life of the enzyme when administered intravenously.Article Acute Abdominal Pain Due To Spontaneous Rupture of the Renal Calyx: Clinical Presentation, Diagnosis and Management(2021) Başok, Erem KaanBackground: Rupture of renal calyces with urinoma, mimicking acute abdomen, is an unusual condition that is mostly caused by distal ureteral stone. Case Presentation: Ultrasonographic evaluation showed right hydronephrosis in a woman who was admitted to our emergency department with persistent severe abdominal pain. A delayed contrast -enhanced computerized tomography (CT) was required to confirm the diagnosis of rupture in symptomatically worsening patient. Among various treatment options conservative treatment was preferred, the patient was recovered, and urinoma was resolved without any complication. Diagnosis and treatment options are discussed. Conclusion: A spontaneous rupture of the urinary system can mimic acute abdomen, and should always be considered in the differential diagnosis of a patient presenting complex symptoms after renal colic. If a definite diagnosis cannot be established by routine imaging modalities, delayed phase contrast- enhanced CT would be helpful in this regard. Conservative treatment with careful monitoring is an option with good results, thus it may obviate the need for surgical intervention in most of the patients.Article Adolescent Health Knowledge of Family Physicians(2023) Aygün, Olgu; Küçükerdem, Halime Seda; Gökdemir, ÖzdenBackground: There are certain life periods when physicians are more cautious with their patients. However, health needs are often disregarded throughout the adolescent era, even though it is a time when fast changes occur in many facets of life, including physical, psychological, cognitive, and social development. This study aimed to contribute to the improvement of adolescent health by measuring the degree of knowledge of family physicians (FPs) on adolescent health services. Material-Method: The was a descriptive-cross-sectional design study. A questionnaire was given to 2200 FPs by e-mail and 344 accepted to participate. The questionnaire had a 13% response rate. Results: Women made up 56.4% of the participants (n=194) and the average age of the participants was 38.77 years (SD: 10.08) (min: 25, max: 80). Two hundred fifteen (62.5%) of the FPs who participated in the survey, had encountered adolescent patients. Methods: Thirty-six Wistar rats were randomly divided into 6 groups based on follow-up periods and administered a dose of adreno-medullin hormone. In each group, bilaterally, a 2-mm bone defect was created at the diaphysis of the radius. Sodium chloride solution was administered to sham groups 3 times a week for 4 and 8 weeks intraperitoneally. Adrenomedullin was administered to the study groups 3 times a week: 15 μg—4 weeks, 15 μg—8 weeks, 30 μg—4 weeks, and 30 μg—8 weeks, respectively. After euthanasia, the segmental defects were evaluated by histomorphometric [new bone area (NBA)] and microtomographic [bone volume (BV), bone surface (BS), and bone mineral density (BMD)] analyses. Results: Although the 4-and 8-week 15 μg administered study groups had higher NBA values than the other study and control groups, the histomorphometric analysis did not reveal any statistical difference between the control and study groups regarding NBA (P >.05). In microtomographic analysis, BV was higher in the 15 μg 4-week group than 30 μg 4-week group (296.9 vs. 208.5, P =.003), and BS was lower in the 30 μg 4-week group than in the 4-week control group (695.5 vs. 1334.7, P =.005), but overall, no significant difference was found between the control and study groups (P >.05). Despite these minor differences in histomorphometric and microtomographic criteria indicating new bone formation, the BMD values of the 15 μg 8-week study group showed a significant increase compared with the control group (P =.001, respectively). Conclusion: Adrenomedullin positively affected BMD at 15 μg, but this study could not show healing in the segmental defect site at different dose regimens. The concept of cost-efficient, much higher speed and longer range applications of micro jet engines was previously addressed such that an existing basic turbojet engine was converted into a single spool turbofan without using additional components of booster and low pressure turbine. Normally, this situation emerges matching problems since two spools are required to adjust the fan speed independently. A simple solution was to use a Continuously Variable Transmission (CVT) gearbox to adjust optimal speed for the fan. As a result, missing of the positive functionality of the booster would lump into the fan root to form a unified low pressure compression system (unified-LPC). Such a unified-LPC demands unique characteristics of having an extreme twist, very high pressure ratio and mass flux at the root section than at the tip section, despite the exact opposite is being enforced due to the wheel speed rise with radius. In light of these challenges, this work aims to investigate detailed aerodynamics of an existing design previously made and reported by the authors. It is shown that, despite the aerodynamic loading contrast throughout the span, the unified-LPC can still have a wide operating range and acceptable off-design aerodynamics. Despite many controversies about how Karagoz came into existence, there is a consensus that it emerged in the 16th century. Figures, as cultural objects, have production methods and techniques interwoven with the doctrines of the past. Today, figure-making for different purposes, inherited from the past, is present in Karagoz figure art. The aesthetic reflections, technique, and material harmony of the figures for show purposes must carry the cultural essence of Karagoz. This study examines museum collections and exhibitions, interviews with Karagoz masters, and a data analysis from an academic perspective. Considering these factors, it becomes clear that the training of masters for figure-making art is significant. Long-term training organized by institutions will contribute to this art and might be beneficial for good examples to reach the audience. Transferring the figures to future generations while preserving their cultural essence, making them in a suitable technique, and aesthetic understanding synthesized with modern techniques; will also contribute to the survival of this art.
